چکیده

This study discusses poly(3-hydroxybutyrate-co-4-hydroxybutyrate) that is produced by Delftia acidovorans bacteria in fed-batch cultures from various carbon sources. The experimentally determined optimal fed-batch culture conditions are as follows: pH: 7.0, incubation temperature: 26 8C, incubation time: 72 h, and concentration of carbon source: 10 g/L. The maximum 4HB unit composition of P(3HBco-4HB) obtained using the fed-batch culture with a cell concentration of about 2.5 g/L was 94 mol% although the P(3HB-co-4HB) content remained low, at around 13 wt%. The extent of the enzymatic degradation of P(3HB-co-4HB) was determined by involved PHB depolymerase and lipase. The tensile strength of P(3HB-co-4HB) markedly decreased during 2 h of degradation by PHB depolymerase, whereas that of P(3HB-co-4HB) degraded by lipase for 50 h was at least 3 MPa. 2008 Taiwan Institute of Chemical Engineers. Genetic analysis of Comamonas acidovorans polyhydroxyalkanoate synthase and factors affecting the incorporation of 4-hydroxybutyrate monomer.

The polyhydroxyalkanoate (PHA) synthase gene of Comamonas acidovorans DS-17 (phaCCa) was cloned by using the synthase gene of Alcaligenes eutrophus as a heterologous hybridization probe. Complete sequencing of a 4.0-kbp SmaI-HindIII (SH40) subfragment revealed the presence of a 1,893-bp PHA synthase coding region which was followed by a 1,182-bp beta-ketothiolase gene (phaACa). Both the transla...

Microbial degradation of poly(3-hydroxybutyrate) and poly(3-hydroxybutyrate-co-3-hydroxyvalerate) in soils.

The microbial degradation of tensile test pieces made of poly(3-hydroxybutyrate) [P(3HB)] or a copolymer of 90% 3-hydroxybutyric acid and 10% 3-hydroxyvaleric acid was studied in soils incubated at a constant temperature of 15, 28, or 40 degrees C for up to 200 days. In addition, hydrolytic degradation in sterile buffer at temperatures ranging from 4 to 55 degrees C was monitored for 98 days. D...

Biochemical and molecular characterization of the polyhydroxybutyrate depolymerase of Comamonas acidovorans YM1609, isolated from freshwater.

Comamonas acidovorans YM1609 secreted a polyhydroxybutyrate (PHB) depolymerase into the culture supernatant when it was cultivated on poly(3-hydroxybutyrate) [P(3HB)] or poly(3-hydroxybutyrate-co-3-hydroxyvalerate) [P(3HB-co-3HV)] as the sole carbon source. The PHB depolymerase was purified from culture supernatant of C. acidovorans by two chromatographic methods, and its molecular mass was det...
